Calcirol contains the secosteroid cholecalciferol, also known as vitamin D3. Aside from its well-known role in calcium metabolism, this secosteroid has very important role in immune function, muscle health, stress adaptation (cortisol), anti-aging, sex steroid modulation, cardiovascular function, cognitive function, etc.

CamphoSal is a liquid product containing a combination of camphoric acid and phenyl salicylate (Salol). Both of these ingredients were widely used clinically in the late 19th and early 20th century as intestinal antiseptics / disinfectants when antibiotics were not widely available and barely known. There is some evidence that both may have beneficial effect beyond modulating the intestinal microbiome (e.g. gonorrhea), however there is no recent research in that area, and as such no definitive statements can be made.

However, unlike the plain saturated fatty acids the esterified versions are resistant to metabolism and as such have much longer half life, which allows them to exert their effects in much lower concentrations. TocoVit is a liquid, mixed tocopherol supplement extracted from wheat germ oil (WGO). As such, in addition to its high proportion of alpha-tocopherol it also contains a number of other ingredients extracted from the WGO that may have hormonal and metabolic effects on their own. Diamant is a liquid product containing the chemical adamantane. Derivatives of adamantane have been in clinical use for decades are known for their anti-viral, anti-bacterial, pro-dopamine, and anti-excitotoxic properties. More recent studies have discovered that adamantane and its derivatives may be powerful inhibitors of HDAC (in nanomolar concentrations: EC50 = ~70 nM/L). Also, admantane (and its derivatives) may be a direct stimulator of the enzyme tyrosine hydroxylase and thus a direct stimulator of endogenous dopamine synthesis. In addition, adamantane and its derivatives have been studied if they inhibit cortisol synthesis, lower plasma serotonin, histamine, blood glucose, insulin, and inflammatory biomarkers. The extreme lipophilicity of adamantane has led to its use as an absorption enhancer for various drugs delivered topically, orally or even IV and this lipophilicity seems to also enhance the half-life of the chemical administered together with adamantane. For example, several derivatives of aspirin, indomethacin and testosterone have been synthesized using adamantane as the modifier and the bioavailability and half life increases from a few hours to as long as 8 weeks (i.e. see the liphophilicity links in the references below; the steroid Bolmantalate is a direct example Bolmantalate - Wikipedia).

Lanosterol is the primary precursor of cholesterol (and thus steroids) of animals and fungi. It has been shown to possess a number of beneficial health effects including reversing protein aggregation in neurodegenerative diseases, pro-metabolic (uncoupler), anti-inflammatory, anti-endotoxin, anti-glucocorticoid, anti-viral, anti-mutagenic, antimicrobial, anti-cancer, anxiolytic, antidepressant, and generally anti-aging.

Lisuride is a chemical agent of the iso-ergoline class, related to the dopaminergic ergoline Parkinson's drugs. Lisuride is a full dopamine and a mixed agonist/antagonist for several serotonin receptors. It is an antagonist at the serotonin 5-HT2B receptor, which is its main mechanism of action in reversing fibrosis of various tissues and organs. It has a high affinity for the dopamine D2, D3 and D4 receptors, as well as D1, and D5. It is an agonist of 5-HT1Aand 5-HT2C receptors, and putative antagonist on 5-HT2A (which explains its antagonism to LSD hallucinogenic effects). Lisuride is also a putative histamine antagonist, on both H1 and H2 receptors.

Metergoline is a non-selective serotonin antagonist and putative dopamine agonist. As a serotonin antagonist it targets the 5-HT1, 5-HT2, 5-HT6, and 5-HT7 receptors but due to its general antiserotonin effects it is considered a non-specific serotonin antagonist similar to cyproheptadine. The dopamine agonism profile of metergoline is not well-known except some evidence for D2 receptor agonism. Metergoline is capable of lowering prolactin through both dopaminergic and non-dopaminergic mechanisms and displays similar effectiveness to bromocriptine in the same dose range, and the two drugs are considered synergistic. One of the most interesting properties of metergoline is its antifungal activity against Candida strains, which are naturally resistant to anti-fungal drugs.

6-ketoprogesterone (6-keto P4) is an orally active oxidized form of progesterone that contains a keto group at position C6. According to studies, it has pronounced anticatabolic (hypertrophic) effects on muscle tissue equal to 2/3 of the effects of testosterone. In addition, the studies claim that 6-keto P4 has no estrogenic, antigonadotropic, thermogenic, or other effects commonly associated with progesterone, while having only about 1/6 of the sedative effects of progesterone. As such, 6-keto P4 effects appear to be focused almost entirely on opposition to cortisol - i.e. antagonism to GR and subsequent downstream effects associated with glucocorticoids.

A naturally occurring (e.g. in tobacco) β-carboline derivative, 10-methoxy-harmalan (10-MeO-Harmalan) has been shown to act as a serotonin antagonist both in-vitro and in-vivo, with a mechanism and potency similar to the ergot derivatives, but without their psychotropic/hallucinogenic effects. In addition, several studies suggest dopaminergic effects of most naturally occurring β-carbolines. The chemical is currently not known to be regulated/restricted and is available commercially from a number of major chemical vendors.

Allopregnanolone is a metabolite of progesterone through the action of the enzymes 5α-reductase (5-AR) and 3α-HSD. Unlike progesterone and 5α-DHP, it is NOT an agonist of the progesterone receptor (PR). However, like progesterone and 5α-DHP, it is an agonist of the GABA receptor and is perhaps the most potent endogenous GABA agonist. Due to its 5-AR derived nature allopregnanolone can serve as a precursor to androgens such as DHT, androstanedione, and androsterone through a recently discovered alternative pathway. Allopregnanolone levels drop in cases of severely downregulated 5-AR activity due to stress, PUFA overload, chronic disease, aging, and administration of 5-AR like finasteride or dutasteride (i.e. the (in)famous Post Finasteride Syndrome (PFS)).

5α-Dihydroprogesterone (5α-DHP) is a metabolite of progesterone through the action of the enzyme 5α-reductase (5-AR). It is an agonist of the progesterone receptor with similar potency to progesterone, and just like progesterone it is an agonist of the GABA receptor. As a result, 5α-DHP has been shown to possess a number of properties ascribed to progesterone, including anti-estrogenic, anti-prolactin, sedative, anxyolitic, anti-depressant, neuprotective, anti-aging, pro-metabolic, pro-thyroid, and anti-proliferative. Unlike progesterone, 5α-DHP is not a metabolic precursor to cortisol, estrogen and androsterone. However, due to its 5-AR derived nature 5α-Dihydroprogesterone (5α-DHP) can serve as a pro-hormone to potent androgens such as DHT, androstanedione, and androsterone through a recently discovered alternative pathway. 5α-DHP is also a direct precursor to allopregnanolone and can elevate its levels even in cases of severely downregulated 5-AR activity due to stress, PUFA overload, or administration of 5-AR like finasteride or dutasteride. Finally, unlike progesterone, 5α-DHP seems to have little or no anti-androgenic effects.

Androsterone is an androgenic steroid derived via the activity of the enzyme 5-AR and is a donwstream metabolite of the more potent androgen DHT. Like all 5-AR derived androgens, androsterone displays anti-estrogenic and anti-glucocorticoid activity and in addition serves as a pro-hormone for DHT and other potent androgens. In addition, androsterone is a neurosteroid with potent GABA agonist activity, and is known to function as a pheromone in many animal species including humans. It has been shown to possess anti-depressant and anti-proliferative effects. Perhaps most importantly, it has been found to act like as a potent thyroid mimetic / surrogate and as such to increase basal temperature, oxygen consumption and lower lipid levels in humans.
